I did assistant manager then the infected ladybug then mant then shmector and that's what I've done so far. I did: infect. ladybug, asst. manager, broodmother, mantis, schmector, and have yet to do the javamatic. I do think I should've just done the director first because mantis is so much harder. Honestly, Shmector was underwhelming to me.

A Small Wasp Nest is a resource node for Wasp Paper, just like Berries or Pupas are resource nodes, so it probably respawns. Don't know how long it take but the nests do respawn after a couple days. I destroyed the one under the Javamatic and it's back.
